Dynamic chamber volume formula

Volume (µL) = Counting area (mm²) × Depth (0.1 mm)

Cells/µL = Cells counted × Dilution factor ÷ Volume (µL)

1 cell/µL = 1 cell/mm³.

Counting area Volume
1 large square 0.1 µL
2 large squares 0.2 µL
4 large squares 0.4 µL
5 large squares 0.5 µL
User-defined area Area × 0.1 µL

Improved Neubauer chamber

Region Use
Corner large squares Four corner large squares — each 1 mm², commonly used for CSF nucleated cell (WBC) counting.
Central large square Central large square — subdivided into 25 smaller squares; often used for RBC/platelet counting in classic hemocytometer protocols.
Additional large squares Remaining large squares — may be counted depending on the protocol or when a larger area is needed for low-cellularity specimens.
Chamber depth = 0.1 mm Chamber depth is fixed at 0.1 mm between the coverslip and the ruled surface — this is what converts a counted area (mm²) into a counted volume (µL).

Module notes

The RBC module is independent from the WBC module — select the chamber region actually used for the RBC count; it is not assumed to match the WBC counting area.

Correction formulas for traumatic CSF contamination are estimates and should not replace clinical correlation, microbiological investigation, or cytological assessment.

A calculated blast percentage does not by itself establish a diagnosis of CNS leukemia or malignancy. Morphological identification and, where appropriate, flow cytometry/cytological evaluation are required.
